چکیده

Accordingly, it was of interest to study argininecanavanine relationships in actively multiplying tumor cells. Arginine has been shown to be essen tial for the growth of the Walker carcinosarcoma 256 in vitro (8) ; and, with the recent developments in tissue culture (9, 11) in this laboratory, it was possible to study the effects of canavanine on argi nine metabolism during tumor cell growth. The present report describes this investigation as well as related growth experiments with ornithine and citrulline.

Incorporation of canavanine into protein of Walker carcinosarcoma 256 cells cultured in vitro.

The inhibitory effect of canavanine on the growth of Walker carcinosarcoma ~56 cells in vitro was described in a previous report (9). Apparently, the inhibition rested in a competitive interference of canavanine with the utilization of arginine. A number of recent reports have demonstrated that certain amino acid analogs--e.g., ethionine (6, 10, ~ , ~6), p-fluorophenylalanine (2, 17, 9.~5), fl-...

The arginine requirement of tissue cultures. I. Interrelationships between arginine and related compounds.

The recent development of synthetic (1, 2) and semisynthetic (3) media that support the survival and propagation of mammalian tissue cultures has made it possible to determine the specific amino acid requirements of many cell lines. A remarkably similar pattern of essential and nonessential amino acids has been found for chick embryonic heart fibroblasts (4), Strain L cells (3), HeLa cells (5))...

The effect of varying implant cell numbers on the inhibitory activity of guinea pig serum on Walker carcinosarcoma 256 in the rat.

An attempt was made to ascertain the period of latency and the percentage of takes of the Walker carcinosarcoma 256 in rats after implantation with different numbers of tumor cells. Implantation of 50,000 cells has been shown to result in tumor growth in Sprague-Dawley and Wistar rats, whereas 10,000 cells proved inadequate for initia tion of tumor growth. The length of the latent period is inv...

Inhibitory effects of D-glucosamine on the growth of Walker 256 carcinosarcoma and on protein, RNA, and DNA synthesis.

Fjelde et a!. (9) observed that D-glucosamine inhibited the growth of human epidermoid carcinoma cells in tissue culture, with concentration of 1 mg/mI causing death of the cells within 24 hr. The i.p. administration of divided doses of D-glucosamine or 2-deoxy-D-glucose inhibited the growth of Walker 256 carcinosarcoma in rats without significant toxicity to the host (1). Tumor growth resumed ...
